Vector Maps

Here are the maps for the AdZ vectors. Plasmid maps are in genbank format - this can be imported by most molecular biology programs. It's a text format which includes both sequence & annotations. They're also here as plain text (Fasta format) and a pdf which lists the positions of some of their features. Note that the vector numbers are different to those in the original publication - the original work used vectors in which the genome was derived from the AdEasy vectors. The new vectors contain a newly derived & fully sequenced Ad5 genome, but the expression cassettes etc are identical to those in the original paper. In addition to these E1/E3 deleted vectors we also have versions that are E3 intact, or fully intact.  The maps for those are at the bottom.

pAdZ5 is the entire Ad5 genome, in a non-self-excising vector.  If you wish to recover virus from this vector, it must first be linearised by PacI or SwaI digestion.  If you want to make a self-excising wildtype Ad vector, then simply replace the expression cassette in pAdZ5-CV5-E3+ (below) with the E1 region from pAdZ5 in a single recombineering step.
